The SpiNXT Fungal DNA Extraction Kit is based on silica membrane spin-column technology for reliable purification of fungal genomic DNA. The protocol employs optimized lysis buffers and enzymatic treatment to effectively disrupt tough fungal cell walls and release nucleic acids. RNase treatment ensures removal of RNA contaminants, allowing isolation of RNA-free genomic DNA. Following lysis, DNA binds selectively to the silica membrane under high-salt conditions. Impurities such as proteins, polysaccharides, and secondary metabolites are removed through successive wash steps. The purified DNA is finally eluted in a low-salt buffer, yielding high-purity fungal genomic DNA compatible with sensitive downstream applications.
